The test kit for diagnostics of H5N1 avian influenza A virus [29.03.2006]
Unprecedented
epizootic among a poultry population caused by a high pathogenic Í5N1 avian
influenza A virus in the winter 2003-2004 in 8 countries of South and South-eastern Asia
and more frequent cases of human infection in Vietnam and Thailand following with
high death-rate has aroused a defensible anxiety that such tendency in avian influenza
evolution (Í5N1 subtype)
could become a beginning of new pandemic of influenza among human population.
Rash spreading of this disease among
poultry for the period from August to December 2005 at the territory of Russia,
Turkey, Croatia, Romania and Ukraine made anxious a scientific community of the
whole world and international organizations WHO, FAO, BWPPP. In January 2006 the
International practical seminar organized by Ministry of Health of Ukraine and USA
Embassy in Ukraine took part in Kyiv with participation of above mentioned organizations
and representatives from participant-countries (Georgia, Azerbaijan, Uzbekistan,
Kazakhstan, Belorussia). The seminar on the subject of geographically spreading
of such virus and of economic losses and also of development of mechanisms of
international interactions on reducing of risks of influenza pandemic among
human population.
Considering an relevance of the given
problem for Ukraine, especially in spring 2006 scientific researchers of the company
Diaproph-Med in collaboration with Government enterprise «STCImmunobiotechnology STC «Institute of Monocrystal» of National Academy of Sciences of Ukraine and Institute of Veterinary Medicine UAAS
beginning from August 2005 have been developing a complex of molecular methods
for indication and identification of Í5N1 avian influenza A virus.
An elaborated test kit «DIA-Real
Avian Influenza» for the detection of RNA of Í5N1 avian influenza À virus in clinical materials basing on the one-stage Real-Time RT-PCR method is an universal
kit for human medicine and veterinary medicine.
The test kit is intended for 48 sample
investigations (controls including).
The
contents of the kit:
1. the
set of reagents for isolation of viral RNA from clinical materials «QIÀamp Viral RNA Mini Kit».
2. the set of reagents for Real-Time RT-PCR No.1 performance – for the performance of I stage of the
detection of avian influenza A virus.
3. the set of reagents for Real-Time RT-PCR No.2 performance – for the performance of ²² stage of the identification of Í5N1 avian
influenza A virus.
The limit of
sensitivity of the kit is 103 copies/μl, the specificity – 100%.
The
elaborated test kit was approved during screening of biological materials of poultry
provided by State scientific control institute of biotechnology and microorganism
strains.
